Hypoxic preconditioning promotes survival of human adipocyte mesenchymal stem cell via expression of prosurvival and proangiogenic biomarkers

Background: Contributing factors for improved survival of human adipocytes mesenchymal stem cells (h-AMSCs) cultured through hypoxia preconditioning, in example apoptosis inhibition involving BCL2 and HSP27 expression, trigger signal expression (VEGF), SCF OCT-4 CD44+ expression. The objective if this study was to explain the mechanism role hypoxic preconditioning optimal duration exposure improve h-AMSCs. Methods: An experimental laboratory explorative (in vitro) with h-AMSCs cultures. This research conducted four stages. First, isolation culture from adipose tissue patients. Second, characterization by phenotype (flowcytometry) CD44+, CD90+ CD45-expression before being pre-conditioned treatment. Third, performed an oxygen concentration 1% 24, 48 72 hours. Fourth, observation tested on VEGF, SCF, OCT-4, BCL2, Flowcytometry apoptotic Tunnel Assay method. Results: result regression test showed that time difference had effect VEGF (p<0.001;?=-0.482) condition also influenced (p<0.001;?=0.774). path analysis (p<0.001; ?=0.985). results effects ?=0.398) precondition affects ?=0.847). Pathway inhibited (p=0.030; ?=-0.442) (p<0,001;?=-0.487). Conclusion: Hypoxic h-AMSC has proven increase HSP27. demonstrated explained existence a new increased cultures (O2 1%) via HSP 27.
